This study conducted two experiments to investigate the influence of advertising repetition and time pressure on mobile advertisement effectiveness. The first experiment examined the effect of advertising repetition in everyday life. SMS advertisements with different repetitions were sent to participants during 6 weeks. The results indicated that it was better to send less than three mobile advertisements each day. The second experiment examined the effect of time pressure in the lab controlled environment. Under high or low time pressure, participants received SMS advertisements while searching information through webpages. Context-aware applications, which consist of a sensor system, a reasoning system and service artifacts such as mobile devices, kiosks and robots, require data from the sensors to be queried on a continuous basis. The smaller the sensing interval and the greater the amount of service time, the more accurate the service, but the more energy is consumed. Thus, use of context-aware applications always involves a trade-off. In this paper, we propose an automatic method of optimizing the level of personalization involving the sensing cycle and service time of a personalized application. The method proposes a quadratic form of total cost curve which demonstrated that the minimum identified value is always the global optimum. The interactions of antibiotic with living cells were studied by lectin conjugated gold nanoparticles(GNPs)based colorimetric assay. Because of the high affinity of lectin for saccharides, the lectin conjugated GNPs are able to employ as indicators for monitoring the antibiotic induced changes of glycosyl complexes. The interactions of a well known antibiotiC., tunicamycin, with two different cell lines, HeLa and SHG-44, were selected to establish this assay.In the presence of tunicamycin, the dose-and time-dependence on the decreasing of binding affinity of lectin conjugated GNPs with living cells were demonstrated by conventional microscopic and UV-Vis spectroscopic studies. The experimental result demonstrates that our approach can be used to identify antibiotic induced expression difference of glycosyl complexes on different cellular surfaces and determine drug activity quantitatively. For further confirming the capability of the GNP-based assay, the system was also studied by confocal laser scanning microscopy(CLSM)and classic flow cytometry(FCM) assay, and satisfactory results were obtained. 相似文献

Label-free and real-time information acquisition of molecular phenotype and its function on living cells plays a significant role in disease diagnosis and drug development. In this paper, SPR phase sensing was applied to monitor the interactions between EGFR antibody, EGFR1, and membrane proteins EGFR on living human gastric cancer BGC823 cells. When 50 μg/mL EGFR1 was added onto the fixed cells chip and the living cells chip, a significant difference in the binding amount could be observed from the immunofluorescence images. Quantitative results were obtained by following SPR detection, which were 722 RU and 438 RU, respectively. On the same living cells chip, SPR detection also showed markedly different results of cellular responses when it was stimulated by EGFR1 at different concentrations, such as adhesion and/or morphology variation, revealing the EGFR1's cytotoxic effect on the BGC823 cells. The results demonstrate SPR phase sensing is capable of real-time detection of molecular interactions and cellular responses on living cells, and suggest that further studies on the mechanism and the technique may allow SPR sensing become a powerful tool not only for the basic research of cell biology, but also for medical diagnosis and drug development. 相似文献
